Title: Identificación de marcadores genéticos predictores de respuesta al tratamiento endocrino en pacientes con cáncer de mama luminal A

Abstract: Breast cancer is the most frequent oncological disease worldwide in the female population. There are different subtypes depending on the receptors present in the tumor tissue. Luminal A has the best prognosis and the lowest risk of relapse. Its treatment is mainly endocrine. This work focuses on these patients: Luminal A breast cancer in endocrine treatment without chemotherapy. The objective is to search for gene markers that predict a better response to treatment. For this purpose, cause-specific survival is assessed according to the level of gene expression. The gene search is first performed by a targeted univariate analysis procedure, where only the influence of a single gene is studied, and multivariate, studying combinations of genes. Subsequently, a prediction of significant variables in the model is developed, by means of Lasso regression, to obtain the combination of genes that provides the greatest distance between the survival curves. Finally, functional analysis of the results identifies significant metabolic pathways. The conclusions obtained show that there are genes related to improved survival in these patients depending on their level of expression: high expression of SCP2, MYO6, PDLIM, U2SURP and low expression of MRPL23, CAD, FBP1 and RBP2. On the other hand, the combination of MRPL23 and MTA3 with low expression level contributes 99.3% survival rate. In the functional analysis, the fructose and mannose metabolism pathway was significant for the set of predictor genes studied.
