Title: Identificació i caracterització de proteïnes i estructures de superfície de Bordetella pertussis com a potencials nous candidats vacunals

Abstract: Among the possible causes involved in the resurgence of whooping cough over the last few years, stands out the adaptation of Bordetella pertussis to the immunity conferred by acellular vaccines (DTPa), which were introduced into our environment at the end of the 90s. That is why, the aim of the work is to understand the antigenic replacement of B. pertussis from an immunological point of view, as well as to carry out a search for new candidates to be considered in future vaccine compositions against pertussis. For this purpose, an exploration of the B. pertussis genome has been carried out in order to identify the pathogenic mechanisms of the microorganism that could be involved in its infection process, characterising them from a structural, functional, localisation and the antigenic point of view. In addition, an immunological characterisation of the allelic variants of the virulence factors of the microorganism included in the DTPa vaccine compositions has been carried out. On the one hand, among the candidates studied, HUVH-BP6, DNT, AC-Hly, BapC, BrkA and Vag8 were the proteins with the most favourable characteristics to be considered as good candidates to be taken into account in future pertussis DTPa vaccine compositions. On the other hand, the observed variability of B-cell and T-cell epitopes between the different allelic forms of the B. pertussis vaccine antigens included in the currently used DTPa vaccine compositions may have favoured the antigenic replacement of the microorganism as a mechanism of adaptation to the immunity conferred by this type of vaccine since its introduction.
