Title: Aproximación genómica y fenotípica al estudio del papel de la cápsula en la virulencia de Vibrio vulnificus

Abstract: Vibrio vulnificus is a zoonotic pathogen that causes the death of its host by septicemia due to the production of a protective capsule. Reversion from capsulated (opaque colonies, O) to non-capsulated (translucent colonies, T) phenotype and vice versa has been described in this species but with a very low frequency. Unravelling the genetic basis of the reversion would allow not only to understand the pathogenesis of this bacterium but also to design control strategies. The aim of this TFM was to elucidate this mechanism by a genomic approach combined with a phenotypic approach using an uncharacterized clinical strain. For this purpose, the genomes of the O and T variants of this strain were sequenced and compared and, at the same time, assays were performed to relate colonial morphology to predicted phenotypes in the species. The results obtained suggest that the strain studied belongs to a new serovar within Lineage 1 in which the reversion of the phenotype from O to T and vice versa is due to regulatory phenomena yet to be determined and not to reversible genetic changes. In this new serotype, the production of the capsule also confers septicemic capacity but, unlike those described, the reversion from T to O increases with the passages in culture medium suggesting that the reversion is due to a transcriptional regulation phenomenon related to adaptation to the environment. Finally, the genome of both variants presents an inovirus whose characterization deserves future studies.
