Title: Análisis del patrón de expresión de la tolerancia a endotoxinas en un modelo de monocitos humanos mediante RNA-seq

Abstract: The aims of this work is to evaluate the response of the Immune System focusing on the Endotoxin Tolerance. This is described as the reprogramming of the innate immune system against infections caused by pathogens and is one of the most important mechanisms of protection described in biomedicine. In this study, three models were developed with human monocytes/macrophages. These models simulate a healthy donor, an inflamed patient and the state of endotoxin tolerance. To carry out a comprehensive differential study of the three groups, we used the RNA-seq technique, since it quantifies the presence of messenger RNA in a biological simple. Moreover we use it to detect changes in the transcriptome between these conditions, thus obtaining specific characteristics and genetic changes in endotoxin tolerance. Once the analysis is developed using various computer tools, we can conclude that the different expression patterns of each model are checked. This fact allows clarifying the independence of each state and obtaining listings of differential expression genes and their associations to biological processes and functions that may be of great interest in future research. We also generated a list of immunocheckpoints associated with the v-set domain by the research group's own.
