Abstract: Acute lymphoblastic leukemia (ALL) cancer is one of the most common with a high mortality rate in childhood. Throughout the years of investigation have been developed therapies such as radiotherapy or chemotherapy. This work focuses on the chemotherapeutic drug metrotrexate (MTX), commonly used for its high efficacy by inhibiting the enzyme Dihydrofolate reductase(DHFR): A protein related to the synthesis of amino acids, purines among other elements. However, there are cases of resistance to the drug. The objective of this project is searching the bibliographic and understanding the mechanisms of resistance and the studying of the interactions of DHFR-MTX through structures crystallized in data sources. One of the challenges has been to find other mutated experimental proteins to calculate and analyze the drug-protein interaction energies, from which to compare them to find common interactions in both cases. Therefore, computational models of the MTX interaction with WT and resistant DHFR have been made, as a basis for search, and studies of new molecules that may have activity with proteins. Because of this search and study of interactions 3 candidate ligands have been proposed, one of them the drug called Leucovorin for its possible application in the treatment of all. Finally, once all the results have been analyzed, it is concluded that, within the ligands tested, MTX is the one with the most favorable interaction energy. In addition, coinciding with expectations, resistant proteins show worse interaction than wild-type DHFR.
