Abstract: Caffeine is an ergogenic aid with huge scientific evidence regarding its efficacy. However, various factors that could cause important interindividual differences in its effect have been established, among which genetics can be highlighted. Consequently, this influence has been investigated focusing on single nucleotide polymorphisms (SNP) in different genes, such as the CYP1A2 or the HTR2A. The main goal of this master's final project was to develop a systematic review of randomized clinical trials that assessed the impact of the SNP -163C>A (rs762551) in the CYP1A2 gene and/or of the SNP T102C (rs6313) in the HTR2A gene on the ergogenic effect of caffeine on physical performance. This was conducted via a search strategy in 4 databases: PubMed, Scopus, Cochrane Library, and Google Scholar. As a result of this procedure, 16 investigations for the CYP1A2 gene SNP and 1 for the HTR2A one were analyzed. Regarding the studies of the CYP1A2 gene SNP, they showed contradictory findings, and even though the majority of them reported no significant impact of it on the ergogenicity of caffeine, few determined greater benefits from it for the AA genotype carriers. With respect to the HTR2A gene SNP, it did not prove significant influence on the impact of caffeine on its own, although the people with CC genotype in it and the AA one in the aforementioned CYP1A2 gene SNP could obtain greater ergogenicity from it. Notwithstanding, the scarcity of studies conducted on these SNPs does not allow to draw definitive conclusions.
