Title: Impacto de la mutación del gen supresor tumoral PTEN en el tratamiento en cáncer

Abstract: Cancer is the second leading cause of death globally. It develops mutations in order to proliferate and one of the most frequent one is the PTEN mutation. Objective, to identify a metabolic/signalling pathway common in various cancers that plays a role in the alteration of PTEN. To identify genes included in these pathways susceptible to be new therapeutic targets. Methodology, microarray databases (GEO data) have been used. Five types of cancers were obtained with PTEN deletion (colon, breast, epidermoid and non-small cell lung cancer in Homo sapiens and prostate cancer in Mus muculus). Different expression between PTEN knock-out and control cancer samples was analysed, and statistically significant signalling pathways were extracted. Genes included in these pathways were analysed. Results. The small cell lung cancer (KEGGID: 05222), the p53 signalling (KEGGID: 04115) and pathways involved in cancer (KEGGID: 05200) were statistically significant pathways in colon, epidermoid and breast cancer, when comparing PTEN knock-out and control. Common genes in these pathways were TRAF4, TP53 and CDKN1A. Conclusions. TRAF4, TP53 and CDKN1A are susceptible to be therapeutic targets in those cancers with PTEN deletion.
