Abstract: The CRISPR-Cas9 tool allows the genomic editing of a specific DNA site in individual cells or complex organisms. The modification of Cas9 nuclease allowed the use of this technique to create different cellular models, animal models, make functional screenings or perform gene therapy in only 5 years since its description. Currently CRISPR is being used in clinical practice, two therapies have already been approved in China and the United States. Due to its boom, the knowledge of this technique is required for any researcher. In this paper we present the types of mutations performed with CRISPR-Cas9, its application in drug discovery, the generation of disease animal models and the latest advances in gene therapy.
