Abstract: Mantle cell lymphoma (MCL) is considered an incurable disease characterized by relapses and diminished response to therapies. Because new drugs are in need, a statistical analysis was conducted to detect differentially expressed genes (DEGs) in patients with MCL compared to healthy individuals in order to identify new molecular targets. Sixty-five DEGs were obtained which were further analyzed by gene set enrichment analysis with DAVID and protein-protein interaction analysis with STRING. The resulting 65-protein network was evaluated using Cytoscape in order to identify key proteins in MCL lymphomagenesis. CCND1, BCL2, CDK4, CDK6, and MMP9 were the most relevant proteins in this study. After bibliographic research, it is proven that therapeutic design efforts against CCND1 have been infructuous whereas those of BCL2 have been successful in the form of the inhibitor venetoclax. As for CDK4/6, an evaluation of the inhibitor palbociclib in MCL is ongoing. Finally, MMP9 is a promising drug target in MCL and the significance of its inhibition with andecaliximab should be assessed in clinical trials.
