Title: Análisis comparativo de genes productores de piRNA en mamíferos

Abstract: PIWI-interacting RNAs (piRNAs) are 21-35 nucleotide long noncoding RNAs. This type of small RNAs are created in specific loci called piRNA clusters (piCs). The function of piRNAs is to repress the propagation of transposons over the genome. To defend the cell against transposons piRNAs guide PIWI proteins to transposons causing their repression (transcriptional repression) or cleaving the transcripts (post-transcriptional repression). Although PIWI proteins are conserved among different species piRNAs are not. In this project we studied the conservation of piRNA produced in protein-coding genes and their repeat content in a set of previously predicted piRNA clusters from 10 different mammals. First, we found that in all mammals, protein-coding genes produce piRNAs from both exons and introns. Second, we analyzed the conservation of those genes that generate piRNAs. Although most mammalian genes are conserved, only a small fraction produce piRNAs in more than one species. Nevertheless, we found a small number of deeply conserved piRNA producing protein coding genes. Last, we analysed repeats in piCs, genic piCs and protein-coding genes. We found that there is higher density of LTR repeats in piC when compared to other protein-coding genes and the intersection of piCs and orthologues piRNA generating genes. We conclude that production of piRNAs from protein-coding genes evolves fast, with notable exceptions, and that the difference in repeat content between genic and intergenic piCs suggests different functions or modes of evolution.
